摘要
目的:研究新生鼠窒息后肾损伤的超微结构变化。方法:将44只7—10日龄Wistar大白鼠随机分为四组,即对照组(13只)、窒息后复氧2h组(10只)、窒息后复氧24h(11只)、窒息后复氧48h组(10只)。制成常压窒息模型。在上述各时点取肾组织用JEM-100透射电镜观察其超微结构变化。结果:窒息后复氧2h可见近曲小管分界不清,基底质膜内折,微绒毛减少,溶酶体消化线粒体,间质中有中性粒细胞浸润,胞浆内线粒体有髓样变。肾小球次级足突有融合,毛细血管内皮细胞略增生。在24—48h,除肾小管有改变外,肾小球的改变更明显:肾小球次级足突有融合,血窦腔内有红细胞、白细胞及破碎的细胞,滤过膜有增生。结论:窒息后复氧可导致肾脏明显超微结构改变.早期以肾小管损伤明显,后期出现肾小球的损伤。
Objective: To study the ultra - structural changes of renal injury in postasphyxial neonatal rats. Methods: 45 neonatal Wistar rats of 7 - 10 days old were randomly devided into four groups:the control (n = 13), 2h post-asphyxia (n= 10), 24h post - asphyxia(n = 11), and 48h post - asphyxia (n = 10). The experiment groups were established by normobaric asphyxia. The tissues of kidney taken at above - mentioned time were observed by JEM - 100 electron microscope. Results: Changes observed in 2h post - phyxia group: unclear demarcation of proximal convuluted tubules, glomerular basement membrane inversion, microvilli decreased, mitochondria digested by lysosome, neutrophil infiltration in ground substance, and pulpiform change of mitochondria in cytosol, the secondary podocyte of renal corpuscle inosculated, and epithelial cells of capillary vessel slightly proliferated. In 24 and 48 post - phyxia groups. The changes of renal corpuscle is more distinct, the secondary podocyte of renal corpuscle inoseulated, there were erythrocytes, leukocytes and broken cells in blood sinusoid, and hyperplasia of filtration membrane. Conclusion: The ultra- structural changes of kidney tissue in postasphyxial neonatal rats is distinct. In early stage the renal tubule injury is remarkable. In later stage, renal corpuscle injury occurred.
