                    采用分光光度法研究了蛋白质与钇(Ⅲ)-对乙酰基偶氮胂配合物的结合反应,确定了反应的最佳条件,建立了以钇(Ⅲ)-对乙酰基偶氮胂配合物为光谱探针,分光光度测定蛋白质含量的新方法。在pH3.7~4.0HAc—NaAc缓冲介质中,蛋白质与钇(Ⅲ)-对乙酰基偶氮胂(AsA—pA—Y(Ⅲ))配合物发生作用,使配合物谜色。当以配合物的最大吸收波长(653nm)为检测波长时,蛋白质(以人血清白蛋白为例)的质量浓度在0—20mg/L范围内与配合物吸光度的降低值,呈现良好的线性关系,其表观摩尔吸光系数ε654=1.98×10^6L·mol^-1·cm^-1。该法简便,快速,灵敏度高,抗干扰能力强,应用于人血清样品中蛋白质总量的测定,结果满意。
                
                A novel method is developed for the spectrophotometric determination of protein by using Yttrium(Ⅲ) -p-acetylarsenazo ( AsA - pA - Y(Ⅲ) ) as the spectral probe. It is based on the fact that the absorbance of AsA -pA -Y(Ⅲ) complex at 653 nm would be reduced through the interaction between AsA -pA -Y(Ⅲ) and protein in HAc -NaAc buffer at pH 3.7 -4.0. The decrease of absorbance of the AsA - pA - Y(Ⅲ) complex is found to be proportional to the concentration of protein in the range of 0 - 20 mg/L for HSA and the apparent molar absorptivity of the AsA - pA - Y (Ⅲ) cornplex at 653 nm is 1.98×10^6L·mol^-1·cm^-1. The method has been applied for the determination of total amounts of protein in the human serum samples with satisfactory results.
