摘要
目的建立HPLC法,同时测定不同产地天麻中天麻素含量。方法采用Diamonsil^TM C18色谱柱(200mm×4.6mm,5μm),流动相为甲醇-水-冰醋酸(体积比为5:95:1),流速为1.0mL·min^-1,检测波长为270nm。结果天麻素质量浓度在20~200mg·L^-1内呈良好的线性关系,平均回收率为98.2%,RSD为2.7%(n=9),4个产地天麻中的天麻素质量百分含量分别为0.381%、0.454%、0.620%、0.332%。结论本法可用于天麻中天麻素的含量测定;样品溶液制备方法比药典方法天麻素提取更完全;4个产地的天麻中天麻素含量有明显差异,四川青川产天麻中天麻素含量最高。
Objective To set up HPLC method for the determination of the content of gastrodine in Rhizoma Gastrodiae from different regions. Methods A DiamonsilTMCl8 column(200 mm × 4.6 mm, 5 μm) was used with the mobile phase consisting of methanol-water-acetic acid( V:V: V = 5:95:1), The flow rate was 1.0 mL·min^-1, and the detection wavelength was set at 270 nm. Results The liner range of gastrodine was 20- 200 mg·L^-1( r = 0. 999 8). The average recovery of gastrodine was 98.2% (RSD = 2.7%, n = 9). Conclusions The method is suitable for determination of gastrodine in Rhizoma Gastrodiae. The results demonstrated that the method are better than that in CP2000. There existed significant differences in the contents of gastrodine in Rhizoma Gastrodiae from four regions. Amoung them, the content of Rhizoma Gastrodiae from Sichuan Qingchuan is the highest.
