The Expression of NOD Associated Proteins in Lungs of Mice Infected with Asper-gillus Fumigatus

摘要 目的:研究核苷酸结合寡聚化结构域(nucleotide-binding oligomerization domain,NOD)相关蛋白在烟曲霉(Af)感染小鼠肺组织中的表达。方法:通过经鼻气道滴人Af孢子的方法建立小鼠感染Af的模型,对照组小鼠经鼻气道内滴人等量的生理盐水。分别在不同时间点观察肺部Af负荷及病理学改变,采用RT-PCR方法检测各时间点的NOD1、NOD2及受体相互作用蛋白2(receptor- interacting protein 2,RIP2)mRNA在肺组织中的表达。结果:1)感染组小鼠肺部Af负荷24h达最高峰,48h迅速下降。2)两组小鼠均表现为支气管周围炎症为主的病理改变,感染组小鼠24 h炎症细胞浸润最显著,48 h炎症细胞浸润减轻。对照组小鼠病理损害明显轻于感染组。3)NOD1、NOD2、RIP2 mRNA表达在接种Af后12 h、24 h、48 h显著升高(P<0.05)。对照组各时间点NOD1、 NOD2、RIP2 mRNA表达无显著差异。结论:NOD蛋白可能为Af的胞浆内模式识别受体(pattern。recognition receptors,PRRs),在抗 Af肺部感染的免疫反应中发挥一定作用。 Objective: To study the expression of nueleotide-binding oligomerization domain (NOD)associated proteins in lungs of mice infected with Aspergillus fumigatus. Methods: The model of mouse infected with Aspergillus fumigatus was established by dropping Aspergillus fumigatus conidia into the nares of each mouse. Equal volume of normal saline was dropped into the nares of each mouse in control group . Aspergillus fumigatus burden, pulmonary histopathology change were observed at different time points. Semiquantitative RT-PCR was also performed to study the expression of NOD1 ,NOD2 and receptor-interacting protein 2( RIP2)mRNA in lungs at different time points. Resuits: 1. The burden of Aspergillus fumigatus in lungs of infection group mice reached peak at 24h, and decreased rapidly at 48h. 2. Mice in both groups showed peribronchitis in pulmonary histopathology. In infection group, inflammatory cells infilitration was most obvious at 24 h, while clearance of inflammatory cells was observed at 48 h. The histopathologie finding in control group mice was much less severe than that in infection group. 3. The expression of NOD1, NOD2 and RIP2 mRNA increase significantly after inoculated with Aspergillus fumigatus conidia at 12h,24h,48h(P〈0. 05). There were no significant changes in expression of NOD1 ,NOD2 and RIP2 mRNA in control group at different time points. Conclusion: NOD proteins may be the cytoplasmic pattern recognition receptors of Aspergillus fumigatus, and play a role in immune defense on pulmonary Aspergillus fumigatus infection.
