应用高效液相色谱测定油菜叶片中硫甙成分与含量 被引量:7

Application of high-performance liquid chromatography for glucosinolate analysis in Brassica leaves

摘要 本文研究建立了高效液相色谱测定油菜叶片中硫甙总量与分量的测定方法.油菜叶片经20h冰冻干燥,用70%甲醇为提取溶剂,超声波提取8min,1mg硫酸酯酶脱硫酸根,离子交换树脂纯化后,应用高效液相色谱进行油菜叶片硫甙组分定性、定量.该方法加标回收率为94%~103%,样品极差为2.29μmol/g,符合ISO9167国际标准要求. HPLC was applied to analyze the profiles and contents of glucosinolates which were extracted from Brassica leaves with 70% methanol solvent by ultrasonic methods after freeze-drying. Results showed that the recoveries ranged from 94% to 103% , the range was 2. 29μmol/g, which accorded with ISO9167 standard. The method was precise, simple and practical for leave glucosinolate determination. It could be widely used in quality and diseases resistance breeding for analysis of glucosinolates contents and profiles in Brassica leaves.
