复方淫羊藿总黄酮对去卵巢大鼠股骨和肝肾功能的影响 被引量:4

Effects of combination of epimedium pubescens flavonoids and diethylstilbestrol on biochemical indices of femur and serum in ovariectomized rats

摘要 目的:观察淫羊藿总黄酮(EF)与己烯雌酚(DES)组成复方淫羊藿总黄酮(CEF)对去卵巢大鼠股骨和肝肾功能的影响。方法:4.5月龄SD大鼠70只,随机分成7组,除假手术组大鼠外,其余行双侧卵巢切除术。假手术组(Sham)和去卵巢组(OVX)给溶剂对照,其余分别为:DES组(22.5μg.kg-1.d-1)、EF组(300mg.kg-1.d-1)、CEF高剂量组(EF300mg.kg-1.d-1+DES22.5μg.kg-1.d-1)、CEF中剂量组(EF150mg.kg.d-1+DES11.25μg.kg-1.d-1)、CEF低剂量组(EF75mg.kg-1.d-1+DES5.625μg.kg-1.d-1),均灌胃给药,连续90d。大鼠处死后测定左侧股骨羟脯氨酸(Hyp)、钙(Ca)、磷(P)含量;同时测定肝肾功能。结果:CEF高、中两个剂量组大鼠股骨重量、骨钙、骨磷含量及Ca/Hyp比值均高于OVX组,CEF高剂量组的作用比单用DES和单用EF均强。DES组大鼠肝脏重量指数和血清碱性磷酸酶均升高,而且明显高于假手术组。CEF3个剂量组大鼠肝脏重量指数均大于OVX组,但肝功能其它指标无变化,而且CEF低剂量组大鼠肝脏重量指数比DES组小。结论:CEF预防去卵巢大鼠股骨无机质的丢失的作用强于DES,而且对肝脏的损害作用比DES小。 OBJECTIVE To determine the effects of combination of Epimedium pubescens flavonoids and diethylstilbestrol (CEF) on femur, liver and kidney in ovariectomized rats. METHODS Seventy 4.5-month-old female Sprague-Dawley rats were randomly divided into 7 groups. Rats in sham group were sham - operated and treated by daily oral gavage with vehicle. Rats in other six groups were bilaterally ovariectomized (OVX) and treated by either daily oral gavage with vehicle, or diethylstilbestrol (DES) at 22.5 g·kg^-1·d^-1 , or EF at 300 mg·kg^-1·d^-1 , or CEF at different doses (combination of Epimedium pubescens flavonoids( EF)300 mg·kg^-1 ·d^-1 and DES 22.5 g·kg^-1·d^-1 , of EF 150 mg·kg^-1·d-1 and DES 11.25 g·kg^-1 ·d^-1 , of EF 75 mg·kg^-1 ·d^-1 and DES 5. 625 g·kg·d^ -1 ) for 90 days. The left femur was collected to determine bone weight, contents of calcium ( Ca), phosphorus ( P ) and hydroxyproline(Hyp). The functions of liver and kidney were measured. RESULTS CEF at high dose and medium dose prevented OVX - induced decrease in contents of Ca and P of femur. The contents of Ca and P of femur in CEF at high dose group were greater than those in DES group and EF group, while the effect of low dose of CEF in femur was similar to that of DES. Serum ALP and index of liver in DES group was significantly greater than that in OVX group and sham group. CEF increased index of liver, but had no effect on other indices compared with OVX group. Index of liver in low dose of CEF group was lower than that in DES group. CONCLUSION The protective effect of CEF on bone mineral loss of femur in OVX rats was greater than that of DES and EF alone, while the effect on liver of CEF was lower than that of DES in OVX rats.
