摘要
目的探讨膜型基质金属蛋白酶-1在人脑胶质瘤中的表达及其与肿瘤生物学行为和预后的关系。方法用免疫组织化学S—P法检测48例人脑胶质瘤组织和10例正常人脑组织中MT1-MMP和MMP2的表达。结果高级别胶质瘤组织(Ⅲ~Ⅳ)中MT1-MMP和MMP2的阳性表达率显著高于低级别胶质瘤组织(Ⅰ~Ⅱ),且两者的表达呈显著正相关性。在单因素生存分析中,MT1-MMP的表达与患者预后不良有关。结论MT1-MMP在恶性胶质瘤组织中高表达,与胶质瘤的进展和侵袭密切相关,可作为胶质瘤恶性表型的有用指标;MT1-MMP的阳性表达与患者的预后不良有关。MT1-MMP可能与MMP2的活化密切相关,直接或间接的促进脑胶质瘤的侵袭。
Objective To investigate the expression of membrane type - 1 matrix metalloproteinase ( MT1 - MMP) in human glioma and its relationship to clinicopathological factors and prognosis of glioma patients, nethoths The expression of MT1 - MMP and MMP2 protein was detected in 48 human glioma tissues and 7 normal human brain tissues with immunohistochemical assay 1 S -P method l. Results The expression of MT1 - MMP and MMP2 in the patients with high - grade glioma was significantly higher than that in the patients with low - grade glioma and normal brain tissues,and the marked correlation was found between MT1 - MMP and MMP2. The expression of MT1 - MMP was a considerable fator in survival univariate analysis. Conclusion MT1 - MMP is overexpressed in malig- nant glioma,especially in glioblastoma tissues ,and may involve in the progression and invasion of glioma . It can be regarded a useful marker for judging the malignant phenotype of glioma. Moreover,expression of MT1 - MMP can provide useful prognostic information. It may be implicated in the activation of pro - MMP2 and facilitate invasion of human glioma directly or indirectly.
