系统性红斑狼疮患者外周血多形核粒细胞磷脂酶A2、5-脂氧化酶和白三烯A4水解酶mRNA的表达

摘要 目的:探讨磷脂酶A2(cPLA2)、5-脂氧化酶(5-LO)和白三烯A4水解酶(LTA4-H)在系统性红斑狼疮(SLE)患者外周血多形核粒细胞(PMNs)中的表达水平,及其在SLE发病机制中的作用及意义。方法:应用反转录(RT)-PCR检测28例活动期SLE患者和25名正常人PMNs中cPLA2、5-LO和LTA4-HmRNA的表达水平。结果:活动期SLE患者cPLA2、5-LO和LTA4-H的阳性表达率与正常对照组相比差异无统计学意义;但活动期SLE患者PMNs中的5-LOmRNA的平均表达水平(0.891±0.263)明显高于正常对照组(0.641±0.138),LTA4-HmRNA的平均表达水平(0.6582±0.2804)也明显高于正常对照组(0.3392±0.0751),二者差异均有显著性(P<0.01);活动期SLE组cPLA2的平均表达水平(0.630±0.268)与正常对照组(0.504±0.192)处于同一表达水平,差异无显著性。结论:活动期SLE患者外周血多形核粒细胞中5-LO和LTA4-HmRNA的表达增加,通过诱导白三烯B4(leukotrieneB4,LTB4)的产生参与SLE的发病过程。为特异性5-LO抑制剂或LTB4受体拮抗剂治疗SLE提供了理论依据。 Objective: To investigate the roles and significance of cytosolic phospholipase A2 (cPLA2),5-1ipoxygenase (5- LO) and LTA4 hydrolase (LTA4-H) expression in the pathogenesis of systemic lupus erythematosus (SLE). Methods: Reverse transeription-polymerase chain reaction (RT-PCR) technique was used to semiquantitatively analyze mRNA expressions of ePLA2, 5-LO and LTA4-H in the peripheral polymorphonuclear leukoeytes (PMNs) from 28 active SLE patients and 25 healthy subjects. Results: Compared with normal controls, there was no statistical difference between the positive rates of ePLA2, 5-LO and LTA4-H expression in SLE patients in the active stage (P〉 0.05, both) and normal controls. The average levels of 5-LO and LTA4-H mRNA expression in active SLE group were significantly higher than those in normal controls respectively (P 〈 0.01). However, there was no significant difference in the mean level of cPLA2 mRNA expression between patients with active SLE and normal controls (P〉 0.05). Condusions: The mRNA expression levels of 5-LO and LTA4-H in active SLE group increase markedly, which may be involved in the pathogenesis of SLE and provide a valuable basis for the clinical use of specific 5-LO inhibitors and leukotriene B4 receptor antagonists.
