摘要
我们在国内首次建立了检测弓形虫病lgG抗体的微孔板法直接凝集试验(DAT)及其改良法(MDAT)。经近5年的研究和应用结果表明:该试验具有较高的敏感性和特异性,并能检出较早期的lgG抗体。DAT与法国的DAT(法-DAT)和日本的胶乳凝集试验(日-LAT)比较,总符合率分别为97.3%和95.5%。DAT与IFA的比较,阳性检出率无显著差异,符合率为93.4~95.9%。MDAT与丹麦的染色试验(DT)和两种间接荧光抗体试验(IFA)比较,阳性检出率无显著差异,总符合率亦达91.8~96.0%。用WHO国际生物标准化实验室提供的标准的抗弓形虫人血清定量测得MDAT的灵敏度为0.09IU/ml,接近于改良前的灵敏度(0.08IU/ml)和DT的灵敏度(0.11IU/ml),而高于国外同类试剂盒的灵敏度。
The DAT was evaluated by comparing it with the original French DAT,Japanese latex agglutination test and the conventional indirect fluorescent antibody test(IFA).High agreements were observed between Chinese DAT and French DAT(97.3%),Between Chinese DAT and Japanese LAT(95.5%).High agreements were qualitatively shown between DAT and IFA, 95.1%,93.4%and 95.9% for sera of commercial rabbits,experimentally infested rabbits and human beings respcctively. The geometric mean titers of DAT were higer than those of IFA. The DAT titers were markedly high as compared with those of IFA on sera from experimentally infected rabbits.The DAT was then modified that uses Formalin-fixed and Fuchsin basic stained tachyzoites antigen.The modified methods was evaluated by comparing it with the dye test and two indirect fluorescent andtibody tests and by detecting its lowest level of antibodies.Results for 71 serum spccimens indicated that good qualitative agreement exits among these tesis(91.8%to 96.0%).The modified method,standardized to the WHO reference. enables detection of antibodies down to a lecel of approximately 0.09 IU/ml.Because of their ease and low cost of application,with high sensitivity, specificity and duplicability,the DAT and its modified method are considered to be useful for mass screening and sero-epidemiological studies of T.gondii infectin.
