摘要
目的研究大鼠弥漫性脑损伤后Homer1蛋白3种亚型表达规律及其意义。方法SD大鼠105只,体重(300±25)g,分为正常组、假手术组和弥漫性脑损伤(DBI)组,DBI组应用Marmarou大鼠DBI模型致伤,假手术组不予以自由落体打击,其余处理同DBI组,分别在损伤后30min及1、6、12、24、72、168h取大鼠主要脑区及核团进行Homer1a、1b/c蛋白分子的免疫组织化学和免疫印迹分析检测。结果两种检测方法均发现损伤组神经组织中Homer1a蛋白自伤后30min起表达,持续至伤后168h,而正常及假手术组未见该蛋白阳性表达,而Homer1b/c于正常组、假手术组及损伤组均可见明确的阳性表达。结论大鼠弥漫性脑损伤后Homer1a蛋白的表达于损伤前后有显著变化,而Homer1b/c于损伤前后无明显变化,提示Homer1a蛋白分子参与了神经元损伤修复过程。
Objective To study the expression of three Homer 1 subtypes in diffuse brain injury (DBI) of rats in vitro. Methods One hundred and five SD rats, with weight of (300±25)g, were randomized to normal, sham-operated and DBI group. The Marmarou rats DBI models were produced in DBI group and the shamoperated group was treated in the same way given to the DBI group except for “free fall” attack. The rats'main brain areas and nuclei were detected for Homer by innnunoblotting 'after injury at 30 min, 1, 6, 12, 24, 72 and 168 h, respectively. Results The Homerla was positive in the DBI group after injury from 30 min to 168 h and there was no expression of Homerla in normal group and sham-operated group. Homerlb/c presented in the normal, sham-operated and the injury group. Conclusion The study shows that there is significant change in Homerla expression before and after injury,but there is no significant change in levels of Homer lb/c, which indicates that Homcrla is involved in the repair of neuronal injury.
