摘要
目的观察碘伏对兔离体培养眼角膜上皮细胞形态结构的影响,并探讨用于眼部消毒的安全浓度。方法采用体外培养的兔角膜上皮细胞单层于生长旺盛期加入不同浓度(250mg/L,500mg/L,1000mg/L)的碘伏,分别在加碘伏的5min,10min,30min和60min置于相差倒置显微镜和透射电镜下观察其形态学变化。结果在保证有效杀菌效果的前提下,浓度为250mg/L的碘伏对角膜上皮细胞无明显损伤,500mg/L的碘伏作用60min时角膜上皮细胞出现轻度损伤,浓度大于500mg/L的碘伏在作用10min时可见角膜上皮细胞明显损伤。结论250mg/L的碘伏不仅能保证消毒效果,还能对角膜上皮细胞无损伤,认为250mg/L的碘伏是眼内消毒的安全有效浓度。
Objective Through monitoring the iodophor(Povidone Iodine) influence on the morphology of cultured rabbit corneal endothelial cell(RCEC), to explore its safe concentration to the local sterilization. Methods Different concentration iodophor (250mg/L,500mg/L and 1000mg/L) was added to the cultured single layer rabbit corneal endothelial cell dish when RCEC was proliferated well, and to observe them morphology changes under inverted phase contrast microscope and transmission electron microscope after cultured 5 min,10min, 30min and 60min, respectively. Results On the bases of achieving the effective sterilization result, when the iodophor is 250mg/L, do not have any perceivable injury to RCEC, while, it is 500mg/L, cultured 60 min, can induce minor injury to RCEC, while its concentration higher 0.5%, only cultured 10 min, can induce manifest injury to RCEC.Conclusions When the iodophor is 250mg/L, not only can achieve the effective sterilization result, but also can not induce any perceivable injury to RCEC, so it is the safe efficiency chroma to be used in intra-optical sterilization.
