摘要
在粒细胞/巨噬细胞集落刺激因子(GM-CSF)存在下,离体培养人骨髓单个核细胞3周,结果显示有大量具破骨细胞特性的多核细胞形成,细胞核3~20余个不等,多数细胞显示抗酒石酸盐酸性磷酸酶强阳性反应,当与牙本质片共育3周,可在牙本质片上形成典型吸收陷窝,并可见具有破骨细胞形态样的细胞,提示此培养条件下有大量破骨细胞形成.
Theauthorsdevelopedaculturesystemforhumanosteoclasts.Itwasbasedontheincu-bationofmononuclearbonemarowcelsforthreeweeksinthepresenceofGM-CSF.Theresultingcelsexhibitedaltheclassiccharacteristicsofosteoclasts.Theyformedatruesyncytiumwithmorethan20nucleipercel.Thecelsandpartofthemononuclearcelsrevealedastrongpositivereactiontotartrate-resistantacidphosphatase.Whenthesecelswereculturedonhumandentineslicesforthreeweeks,typicalresorptionpitswereformed.Theauthorsbelievethatthereisthepossibilitytoculturelargenumberofosteoclastunderthisculturecondition.
