摘要
目的研究纤维蛋白原(fibrinogen,Fg)Bβ链-854G/A、Bcl-1基因多态性与脑梗死类型及其危险因素Fg浓度、分子功能等的关系。方法采用病例对照研究,应用聚合酶链式反应限制性片段长度多态法(polymerasechain reaction-restriction fragment length polymorphism,PCR-RFLP),检测33例脑动脉主干支梗死者(MCI组)、43例脑动脉穿通支梗死者(PCI组)和74例正常对照组的FgB-β854G/A、Bcl-1 B1/B2基因多态性;应用辅助血浆纤维蛋白原活性测定系统测定Fg浓度、纤维蛋白单体聚合速率与最大光密度的比值(FMPV/Amax)。结果MCI组FgBβ-854A等位基因频率(24.2%)高于对照组(12.2%),PCI组FgBβBcl-1B1等位基因频率(83.7%)高于对照组(62.8%),PCI组FgBβBcl-1B1B1基因型频率(69.8%)高于对照组(41.9%),差异均有统计学意义(P<0.01);FgB-β854GA+AA基因型人群Fg血浆浓度及FMPV/AMAX高于GG基因型人群,但两者比较差异无统计学意义(P>0.05);FgBβBcl-1的B1B2+B2B2基因型人群Fg血浆浓度及FMPV/AMAX高于B1B1基因型人群,两者比较差异亦无统计学意义(P>0.05)。结论FgB-β854位点A等位基因和Bcl-1位点B1等位基因分别是MCI和PCI的遗传易感因素;Bcl-1的B1B1基因型人群是PCI的易感人群;FgB-β854、Bcl-1基因多态性与血浆Fg浓度及其分子功能无明显相关性。
Objective To study the association of fibrinogen(Fg) gene-854 G/A, Bcl-IB1/B2 polymorphisms with the type of cerebral infarction(CI) and the risk factors - Fg concentration, molecular reactivity and others. Methods A case-control study was conducted: 33 patients were cerebral arterial main trunk infarction(MCI) and 43 patients cerebral penetrating arterial infarction(PCI), 74 normals as controls; the FgBβ gene-854 G/A, Bcl-1 B1/B2 polymorphisms were determined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) ; the Fg concentration, FMPV/Amax were tested with assisting plasm Fg activity assay system. Results Rates of FgBβ-854 A alleles in MCI group were significantly higher than those in control group (24.2 % vs 12.2 %, P 〈0.01) and FgBβ Bcl-1 B1 alleles in PCI group were significantly higher than those in control group, too(83.7% vs 62.8%, P d0.01). In PCI group, rate of FgBβBcl-1B1B2 genotype was higher than that of controls(69.8% vs 41.9%, P 0.01). Concentrations of Fg and FMPV/AMAX in GA+ AA group were higher than those in GG group, but had no significance( P 〉0.05). Concentrations of Fg and FMPV/AMAX in B1B2 +B2 B2 group were higher than those in B1 B1 group, but had no significance ( P 〉 0.05). Conclusion The A allele of FgBβ-854 and B1 allele of FgBβBcl-1 were parting the inherited amiable factor of MCI or PCI. The population carrying B1 B1 genotype of Bcl-1 suffered PCI easily. No obvious correlation between beta-Fg gene-854 G/A, Bcl-1 polymorphisms and Fg concentration and its molecular function was observed.
