摘要
应用单卵囊分离技术,获得猪等孢球虫纯种卵囊,给6头3日龄长大二元仔猪每头经口一次人工感染30个纯种猪等孢球虫孢子化卵囊,收集纯种后代,再对6头3日龄长大二元仔猪每头人工感染2 000个孢子化卵囊继代扩增.观察人工感染仔猪的排卵囊规律包括潜隐期、显露期、排卵囊高峰期及每克粪便中卵囊数(OPG值)和仔猪感染后的临床症状,进行了扩增卵囊的孢子化过程及形态结构观察和显微照像.结果表明,感染30个猪等孢球虫孢子化卵囊的仔猪最早在感染后第8d(8 days post-inoculation,DPI 8)从粪中检出卵囊,排卵囊高峰期在DPI 17~21,持续4~5 d,显露期15~18 d;感染2 000个猪等孢球虫孢子化卵囊的仔猪最早在DPI 5从粪中检出卵囊,排卵囊高峰期在DPI 8~13,持续4~6 d,显露期7~13 d.潜隐期、显露期、OPG值与感染剂量存在相关关系,即感染剂量越小,潜隐期越长,显露期越长,OPG值越小;反之亦然.感染30个卵囊的仔猪无临床可见病征;感染2000个卵囊的仔猪呈现腹泻甚至黄色水样腹泻等明显症状.常规培养新收集的猪等孢球虫卵囊,2 h时胚孢子开始分裂,8 h分裂成2团,12 h形成2个孢子囊,20h开始形成子孢子并可见内残体,最早26 h卵囊完全孢子化;72 h时孢子化率达50%,96 h达80%.本试验扩增和继代扩增所获球虫卵囊,与亲本和文献报道的猪等孢球虫生物学特性相一致.
Pure Isospora suis oocysts were obtained through separation and were orally inoculated to six 3-day-old piglets (30 sporuled oocysts each). Oocysts of the next generation were collected and then applied to another six 3-day-old piglets(2 000 sporuled oocysts each) to amplify pure oocysts of Isospora suis. The regularity of oocyst excretion, including the prepatent period,the patent period,the peak period,the number of oocyst in per gram feces(OPG value), clinic symptoms, the sporulate rate and sporulate process, and the shape of oocysts amplified were observed. The results show that in the group of 30-sporuled-oocyst inoculation, oocysts were detected in the feces at 8 days post-inoculation (DPI 8) ,the peak period started at DPI 17 - 21 lasting 4 - 5 days,and the patent period lasted 15 - 18 days;in 2000-oocyst-inoculation group, however,oocysts were detected in the feces at DPI 5, the peak period started at DPI 8 - 13 lasting 4- 6 days, and the patent period lasted 7-13 days. This indicates that the prepatent period, the patent period and the OPG value is correlated with the number of oocyst, namely, the smaller the number of oocyst, the longer the prepatent period, the longer the patent period and the smaller the infection intensity; and vice versa. During the course of infection, the piglets inoculated with 30 oocysts symptoms or developed diarrhea. Being cultured in did not present any symptoms, whilst the other ones showed slight conventional condition, the sporoblast began to divide at 2 h and divided into 2 agglomerates at 8 h. 2 sporocysts appeared at 12 h, sporozoites and residual bodies began to form in the sporocyst at 20 h, and pioneer sporulated oocyst were observed at 26 h. The sporulating rate was 50 % at 72 h and 80 % at 96 h. The biological characteristics of Isospora suis oocyst amplified were coherent to those of the parent species and those reported.
