摘要
目的探讨用于三维重建的颞骨不脱钙连续大薄切片制作方法。方法取新鲜颞骨标本块,不脱钙聚合物包埋,骨切片机连续切片,片厚50μm。结果制片周期短,切片图像清晰,变形小,颞骨内各结构完整。结论颞骨不脱钙连续薄切片制作方法较火棉胶包埋切片方法快捷,切片变形小,且连续切片的层间距小,适合于对颞骨众多细小结构进行准确三维重建的需要。(中国眼耳鼻喉科杂志,2005,5:382~383)
Purpose To explore the new technique of serially sectioning of undecalcified polymer-embedded temporal bone for three-dimensional reconstruction. Methods One fresh temporal bone without any otologic disease was obtained. After fixation and dehydration, the specimen was embedded in methylmethacrylate (MMA) until the MMA was fully polymerized.This temporal bone block was then sectioned horizontally into serial 50-micron thickness using a heavy-duty sliding microtome. Results It took approximately one month to complete the whole procedure and the new technique provided less-distorted, clear and sufficient microanatomical information. Conclusion The undecalcified polymer-embedded temporal bone specimen processing takes less time than the decalcified celloidin-embedded specimen processing. With application of this technique, the serial, less-distorted and less-shrunk slices with little interlayer space are suitable for reconstructing more accurate and detailed temporal bone three-dimensional data.
