摘要
采用热水浸提法,探讨浸提时间、温度、固液比以及浸提次数等因素对金针菇多糖提取率的影响,通过正交实验得到金针菇多糖的优化浸提条件:95℃、固液比为1:30、浸提3h、浸提2次,提取率达1.54%。5%三氯乙酸正丁醇及氯仿正丁醇溶液(5:1)纯化多糖。
The optimal extraction conditions of polysaccharide from Flammulina velutipes were investigated. The extraction temperature, time, frequency, and the proportion of water volume were discussed with the orthogonal test. The results showed that the optimal condition of extraction was adding 30 times of water as much as fruiting body, 95 ~C for 3 hours and extracting 2 times, the yield rate of extraction of crude polysaccharides was 1.54%. The polysaccharides were primary purified with 5% trichloroacetic acid Butanol ag, Chloroform Butanol ag (5:1).
