摘要
目的探讨液氮冷冻对成骨细胞免疫原性的影响。方法体外培养成骨细胞,传代鉴定后液氮冻存3个月,复苏。建立成骨细胞、淋巴细胞混合培养体系(MLC),测定液氮冷冻后成骨细胞的免疫原性。结果体外培养细胞经鉴定为成骨细胞,液氮冻存3个月后存活率>90%。MLC结果显示冻存前后刺激指数(SI)差异有显著性(P<0.01),提示液氮冷冻后成骨细胞的免疫原性降低。结论液氮冷冻是一种理想的降低成骨细胞免疫原性的方法。
Objective To investigate the effects of fluid nitrogen cryopreservation on immunogenicity of osteoblasts. Methods The osteoblasts were cultured and examined by alkaline phosphatase(ALP) stains and calvaria stains, and then cryopreserved for 3 months. By establishing osteoblast and lymphocyte mixed culture model(MLC), the immunogenicity of osteoblasts was examined. Results Cells cultured in vitro were identified as osteoblasts. The differences of immunogenicity of two groups between precryopreservation and postcryopreservation were significant( P 〈 0. 01 ). Conclusion Fluid nitrogen cryopreservation is an ideal method to reduce the immunogenicity of osteoblasts.
