摘要
目的建立猪胆粉及其制品中猪去氧胆酸的含量测定方法。方法以KromasilC18(4.6mm×250mm,5μm)色谱柱为固定相,柱温25℃,以乙腈0.1%乙酸水溶液(50∶50)为流动相,流速1.0mL·min-1;蒸发光散射检测器检测,漂移管温度40℃,以氮气为载气,压力2.0bar。结果猪去氧胆酸在5.4μg~27.0μg范围内线性关系良好,r=0.9994,加样回收率为97.0%,RSD=2.7%(n=5)。结论该方法简便、准确、重复性好。
Objective: To develop a method for the determination of hyodeoxycholic acid in Porcine Gall Powder. Method:Chromatography was performed on a 5μm Kromasil C18 (4.6mm × 250 mm) at 25℃. The mobile phase was composed of acetonitrile -0.1% acetic acid solution (50:50)with a flow rate of 1.0mL·min^-1. A Sedex 55 evaporative light-scattering detector was used with tube temperature of 40℃ and gas pressure of 2.0 bar. Result:A good relationship between peak area and amount of injection Was noted at 5.4μg~27.0μg. Average recovery of hyodeoxycholic acid was 97.0% and the RSD was 2.7%. Conclusion:It was proved that the method was reliable, simple and accurate.
