摘要
本文采用差速离心法分离提纯葱病毒并对其特性进行研究.病毒粒子为弯线性,平均长度728um,直径约15nm,紫外吸收曲线最高峰在256~260nm之间,最低峰为238nm,分子量为27000道尔顿。光学显微镜下可观察到病叶表皮细胞中的不规则内含物,超薄切片电镜观察,可见病毒风轮状、束状内含体。该病毒引起叶绿体、核膜损伤和解体,其寄主范围较窄。
The results obtained from electron microscopic. observation show that Welsh-onion yellow stripe virus (WoYSV) particle is approximately 728um in length, andmost of them are between 600um and 700um. The purfied virus has a typical UV absorption spectrum max. at 256-260um and min. at 238um respectively. Its average molecularweight is about 27000 Dalion. Pinwheel-like inclusion bodies and destroyed chloroplasts have been found in the infected-leaf cells. Under microscope a number of contentsare observed in the infected cells, including round and elipse ones. They have looserstructure compared with nucleus. Some contents nearby the nucleus form comet-likecomplex bodies with nucleus. The host range of WoYSV is narrow.
