摘要
目的为简便快速地进行抗 HBc IgG 和 IgM 测定。方法用抗人 IgM 或抗人 IgG 抗体包被硝酸纤维素膜,用胶体金标记抗 HBc 单克隆抗体,以重组 HBcAg 溶液作为标本中抗 HBc 与金标记抗 HBc 的联接剂,建立了滴金免疫法(滴金法)。结果经与酶免疫法对比测定,二法抗 HBc IgM 符合率为97.2%,抗 HBc IgG 符合率为95.8%。另以相同模式在同一硝酸纤维素膜上分别点加抗人 IgM和抗人 IgG 抗体,同步检测抗 HBc IgM 和 IgG,结果与分别检测的相同。结论该法快速简便,适用于各级医疗单位,有推广应用价值。
Objective To make the determination of anti-HBc IgM and IgG more simple and rapid,Methods A dot immunogo]d filtration assay(DIGFA)has been developed by coating anti-hu- man IgM and anti-human IgG on nitrocellulose(NC)membrane,labelling anti-HBc monoclonal anti- body with colloidal gold and using recombinant HBcAg as linking agent between anti-HBc in the sample and gold labelled anti-HBc.Results The coincidence rate of the results of simultaneous determination of serum samples with enzyme immunoassay(EIA)and DIGFA was 97.2% for anti-HBc IgM and 95.8% for anti-HBc IgG.Simultaneous determination of anti-HBc IgM and IgG on the same DIGFA device by coating separate dot of anti-human IgM and anti-human IgG on the same membrane showed the same results as determined separately.Conclusions DIGFA is more simple and rapid,it is applica- ble to medical laboratories of different levels.Its extension for practical use is valuble.
