摘要
目的探讨不同抗生素在不同时段对创伤弧菌(Vv)感染小鼠心肌的保护作用及观察小鼠心肌超微结构的动态变化。方法将100只清洁级ICR小鼠分为保护Ⅰ组(所用药物含氧氟沙星、异丙嗪、地塞米松等);保护Ⅱ组(所用药物含氧氟沙星、异丙嗪等);保护Ⅲ组(所用药物含亚胺培南、异丙嗪、地塞米松等);保护Ⅳ组(所用药物含克林霉素、异丙嗪、地塞米松等)和阳性对照Ⅴ组,每组20只。Ⅰ~Ⅴ组每只小鼠接种0.2mlVv菌液(约4.4×106菌群数ml),1h后Ⅰ~Ⅳ组肌肉注射相应药物。接着在3、6、12h取各组小鼠心肌组织进行超微结构观察并比较。结果含有该菌敏感抗生素的Ⅰ~Ⅲ组在接种Vv后8h及24h小鼠存活数均显著高于Ⅴ组和Ⅳ组(P<0.01);Ⅰ~Ⅲ组心肌的超微结构损伤较之Ⅴ组和Ⅳ组有较显著的改善,而Ⅳ组小鼠的心肌损伤与Ⅴ组接近。结论敏感抗生素氧氟沙星或亚胺培南与异丙嗪以及地塞米松等联合应用能提高Vv感染后小鼠的生存率,并改善其心肌的损害。
Objective To discuss the protection of different antibiotics for the cardiac muscle of the mice infected with Vv during the different period of time, and to observe the dynamic changer of the cardiac muscle ultrastrueture, Methods To divide 100 IRC mice into five groups: protective Ⅰ group (including ofloxacin, promethazine,dexamethasone, etc.); protective Ⅱ group (including ofloxacin, promethazine,etc.);protective Ⅲ group (including imipenem, promethazine, dexamethasone, etc,); protective Ⅳ group (including clindamycin , promethazine, dexamethasone, etc ) and positive control Ⅴ group . Each mouse of protective groups and positive control group was inoculated with 0.2ml Vv bacteria liquid (about 4.4×10^6FU/ml), after 1 hour, intramuscular injection with the protective drug and then to observe and compare cardiac muscle ultrastructure in the period of 3h 6h,12h, Results after inoculated with Vv for gh or 24h, the of survival of mice inoculated with bacteria sensitive antibiotic was found much more than that of positive control group and clindamycin group (P〈0,01),The injuries of the cardiac muscle uhrastructure were markedly better than those of positive control group and clindamycin group, while the injuries of mice cardiac muscle were similar with positive control group, Conclusion The sensitive antibiotic, such as ofloxacina, imipenem combinded with dexamethasone and promethazine, can increase the survival rate of the mice infected with Vv and improve the cardiac muscle injuries.
