抗坏血酸对IL-1β体外诱导中脑神经干细胞分化为酪氨酸羟化酶阳性神经元作用的实验研究 被引量:4

EFFECTS OF ASCROID ACID ON IN VITRO IL-1β INDUCED DIFFERENTIATION OF MESENCEPHALIC NEURAL STEM CELLS INTO TH POSITIVE NEURONS

摘要 目的体外观察抗坏血酸(ascroidacid,AA)对IL-1β诱导神经干细胞向酪氨酸羟化酶(tyrosinehydroxy-lase,TH)阳性神经元分化的作用,为帕金森病的细胞移植治疗提供实验资料。方法体外培养中脑神经干细胞并传代分组,在有血清条件下分别予以IL-1β及AA+IL-1β诱导分化,TH免疫细胞化学鉴定分化结果并细胞计数阳性比例。结果有血清条件下IL-1β、AA+IL-1β诱导中脑神经干细胞分化为TH阳性神经元比例约为12·6%和14·3%,明显高于对照组(约2·6%,P<0·01);两实验组间相对无统计学差异(P>0·05);联合诱导组TH阳性神经元细胞形态更成熟。结论IL-1β可以明显促进中脑神经干细胞分化为TH阳性神经元,AA能够加强IL-1β诱导的TH阳性细胞的表型成熟,但在我们的实验条件下二者联合未能增加TH阳性细胞诱导率。 Objective To investigate the effects of ascroid acid (AA) on the differentiation of mesencephalic neural stem cells (M-NSCs) into TH-positive (TH-ir) neurons induced by interleukin-1 bate (IL-1β) in vitro. Methods M-NSCs were cultured and passaged, then IL-1β and AA were used to induce differentiation in the presence of fetal bovine serum (FBS). Immunocytochemical techniques and cell-count assessment were used to detect the TH-ir cells differenciated from NSCs. Results The proportion of MNSCs differentiated into TH-ir neurons was about 12. 6% and 14.3% in the IL-1β group and the AA+IL-1β group respectively, which was much higer than that in the control group (2. 6%, P〈0. 01). There were no statistical differences between the IL-1β group and AA+IL-1β group, but the morphology of THir neurons were more mature in the latter than in the former group. Conclusion IL-1β can significantly increase the differentiation of TH-ir neurons. AA+IL-1β can enhance the mature phenotype of TH-ir cells, but the combination did not increase the rate of production of TH-ir cells under our experimental conditions.
