摘要 [目的]探讨青稞提取液抗氧化和脂质过氧化的作用。[方法]对青稞进行酶液的提取,采用蛋白浓度的测定、酶活力的测定、二苯代苦味酰基自由基(DPPH.)法抗氧化活性的测定、组织匀浆MDA比色法测定抑制小鼠离体组织过氧化能力、体外组织脂质过氧化测定。[结果]对青稞粗酶液对DPPH.的EC50为65.47mg/L,清除率低于同浓度的Vc和茶多酚、柠檬酸、Vc与粗酶液均有协同效应,相对而言,Vc的增效优于柠檬酸。青稞超氧化物歧化酶(SOD)酶液对DPPH.的EC50为12.6mg/L,优于茶多酚,与维生素C非常接近。青稞SOD酶液对小鼠离体肝、肾和心组织中的MDA生成均有抑制作用,其中对离体肝组织中MDA生成的抑制效果最明显,浓度为0.25mg/ml时,抑制率为78.9%;此后再增大浓度,抑制率的增加不高,心和肾效果次之。[结论]青稞粗SOD酶液和青稞SOD酶液都有一定清除DPPH.的能力,青稞SOD酶液有抑制脂质过氧化能力。 [Objective] Researching the effect of the antioxidants and lipid peroxidation from Naked Barley. [Methods] Abstracting the crude Naked Barley by means of mensurating the protein concenfration, enzyme activity and the antioxidant actioity by DPPH method, the mensuration of the inhibitory of tissue departing from the body peroxide activity by homogenate MDA cderimetric method and that of the lipid peroxidation of tissnes. [ Results] According to the 50% scavenging concentration EC50 on DPPH, the EC50 of the crude Naked Barley on DPPH was 64.57 mg/L. The scavenging rate of the crude Naked Barley was lower than ascorbic acid and tea polyphenol of the same concentration. Meanwhile when citric acid and ascorbic acid were used together with Naked Barley, ascorbic acid showed stronger synergistic antioxidant activities with Naked Barley. The EC50 of the Naked Barley on DPPH was 12.6 mg/L, lower than tea polyphenol and close to ascorbic acid. And the Naked Barley had obvious effect on inhibiting tissue homogenate MDA. By comparison of the inhibitory rate of tissue homogenate MDA of heart, liver and kidney, it was found that its effect on liver was the best with inhibitory rate 78.9% when the Naked Barley concentration was 0.25 mg/ml. [Conclusion] The extract of 95% ethanol, the crude Naked Barley and the Naked Barley all had effects on scavenging DPPH free radical and inhibiting lipid peroxidation.
