摘要
目的:探讨碱性成纤维细胞生长因子(basicfibro-blastgrowthfacto,rbFGF)抑制鸡形觉剥夺性近视眼(formdeprivatiomnyopia,FDM)的作用及机制。方法:半透明眼罩遮盖法建立90只鸡FDM模型,随机分成6组:正常对照组(A),FDM组(B),FDM+PBS组(C),FDM+bFGF100ng组(D),FDM+bFGF500ng(E),FDM+bFGF1000ng组(F)。按预定时间把bFGF注入小鸡玻璃体腔。眼罩遮盖15d后检影验光,游标卡尺测眼轴长度,RT-PCR检测后极部巩膜基质金属蛋白酶-2(ma trixmetalloproteinase-,2MMP-2)的表达。结果:与A组比较,B组眼轴延长,近视形成,后极部巩膜MMP-2mRNA表达升高。玻璃体腔注射bFGF能明显抑制FDM形成,且下调后极部巩膜MMP-2mRNA表达。随剂量加大,bFGF抑制作用增强,1000ng bFGF能完全抑制小鸡FDM形成。结论:玻璃体腔注射bFGF能通过下调后极部巩膜MMP-2mRNA表达,有效抑制鸡FDM形成。
AIM: To investigate the effect of basic fibroblast growth factor (bFGF) in preventing form deprivation myopia (FDM) in chickens and its mechanism. METHODS: Chicken myopia was induced by translucent goggles. Ninety chickens were divided into 6 groups randomly: normal control (Group A), FDM (Group B), FDM+PBS (Group C), FDM+bF-GFIOOng (Group D), FDM+bFGFS00ng (Group E), FDM+ bFGFI 000rig (Group F). BFGF was injected into chicken vitreous cavities. After myopia was confirmed by optometry, axial lengths were measured at 15d after the covering of translucent goggles. The expression of MMP-2 mRNA in posterior sclera was detected with the reverse transcriptionase-polymerase chain reaction (RT-PCR). ~ RESULTS: Ocular diopter and axial length in Group B increased markedly in comparison with Group A. Moreover, there was the upregulation of MMP-2 mRNA in posterior sclera in Group B. Intravitreal injection of bFGF could prevent FDM and downregulate the expression of MMP-2 mRNA in posterior sciera. The inhibition of bFGF on FDM was a dose-dependent manner. 1000ng bFGF could completely prevent FDM in chickens. CONCLUSION: Intravitreal injection of bFGF could prevent FDM in chick by downregulating the expression of MMP-2mRNA in posterior sclera.
