摘要
Nosemabombycis(N.b)抗血清常受非特异性干扰而难于实用。本试验通过硫酸铵盐析、透析袋及SephadexG—25柱层析脱盐、DEAE纤维素柱层析、组织粉吸收等一系列纯化过程,并经PACE电泳检查表明所获得的高纯度抗体蛋白成分具有很强的特异性。用以制成致敏炭素检液,对Nosemabombycis发生阳性反应,对SCM6、SCM7均不发生阳性凝集反应。经万余张原种实用性试验,证明其灵敏度增强50倍,误判率为0,可以准确无误地检出Nosemabombycis,具有广泛的应用前景。
Lots of studies in serology of Nosma bobyris showed that the antiserum of Nosema bombycis was frequently interfered by its non-specificity and was diffiedt to be put into practical application. This trial removed the non-specificly of antibody protein throrgh a series of purification Processes including (NH4)2 SO4 salting out, dialysis, Sephadex G-25 column chromatography, DEAE cellulose column chromatogrsphy and tissue powder absorption. Examination of PACE, electrophoresis revealed that the highly purified sntibody protein had high specificity. With this antiserum,N.br-IgG sensitized carbon detector was prepared. It showed POsitive reaction to Nosema bombyris spore snd negative reaction to SCM6. and SCM7 spores. Practical examination for over 10000 pieces of Parent eggs confirmed that its sensitivity ass 50 times higher than microscopic diagnosis with no error,indicating that it might be used for detecting Nosema bombycis and its wide applicable prospect.
