核因子-κB在角膜移植排斥反应中作用的初步实验研究 被引量:3

摘要 目的:研究核因子-κB在角膜移植排斥反应中的作用,为进一步研究角膜移植排斥反应发病机制及治疗提供新的思路。方法:建立大鼠穿透性角膜移植动物模型,用免疫组织化学染色法检测角膜移植术后植片中核因子-κB表达,并以植片排斥反应指数(RI)及病理学表现作参照。结果:同基因移植组角膜植片中的角膜基质细胞及内皮细胞有核因子-κB的微弱表达;同种异体移植组角膜上皮细胞、内皮细胞、基质层细胞及新生血管内皮细胞核因子-κB表达增强。各组间核因子-κB的表达强度差异有显著性(P<0.01)。且核因子-κB的表达强度与排斥反应指数(RI)具有明显的正相关性(r=0.883,P<0.01)。结论:核因子-κB可能通过调控免疫和炎症相关基因的表达,参与角膜移植排斥反应的发生,在角膜移植免疫中起着中心调控者的作用。 Objective To investigate the expression of NF-κB in corneal graft rejective reaction in rats. Methods The model of orthotopical corneal transplantation was developed in rats. The rats were divided into two groups: syngeneic control (Wistar to Wistar) and allograft group (Wistar to SD). Immunohistochemistry staining was used to analyze NF-κB expression of corneal graft in each group. Results NF-κB was weakly expressed on keratocytes, corneal endothelial ceils and not expressed on corneal endothelial ceils after transplantation in syngeneic group. NF-κB was obviously expressed on corneal epithelial cells, keratocytes, corneal and vascular endothelial ceils in ailograft group. The difference of NF-κB expression between two groups was significant(P〈0.01). Clinical observation showed the expression level of NF-κB was significantly correlated with rejection index(RI) (r=0. 883, P 〈0.01) . Conclusion NF-κB may regulate the expression of genes associated with graft rejection, and thus play a role of center-control in the occurance and development of corneal graft rejective reaction.
