摘要
将体外培养的大鼠血管内皮细胞(VEC)在不加任何低温保护剂的条件下,以1℃/min的速率分别冷冻至0、-10、-20和-40℃;以10℃/min的速率冻至-40℃,在冻后1d和3d观察VEC生长及其某些功能的改变。结果显示,冻后1d,以1℃/min冷冻的各组VEC数及生长率降低、培养液中乳酸脱氢酶(LDH)活性增加,因子Ⅶ相关抗原(ⅦR:Ag)免疫荧光阳性细胞减少,且冷冻温度愈低,其改变程度愈严重,10℃/min冻至-40℃,上述指标的改变相当于1℃/min的速率冻至-20℃时的情况。冻后3d,各项指标有不同程度的恢复。结果表明,冷冻速率和冷冻温度均与细胞的损伤程度有密切关系。
In order to investigate the injurious effects of cooling rate and degrees of low temperature on endothelial cells(VEC), the present experiments were performed in vitro. The cultured VEC of rat aorta were subjected to Lowering of temperature without any eryoprotector to 0, 10, -20,and -40℃,respectively, at a rate of 1℃/min, and to - 40℃ at 10℃/rain in a cooling apparatus. The frozen VEC were thawed immediately in 37℃ and then recultured. The number and growth rate of VEC, Lactic dehydrogenase (LDH) activity and factor Ⅷ related antigen (Ⅷ R:Ag) in cultured fluid of VEC were determined on 1 and 3 days after freezing by means of ultraviolot. The results showed that the number and growth rate of VEC were lowered, the LDH activity increased, the number and positive rate of cells with immunofiuretcence decreased in all the cell cultured which were frozen at a rate of 1℃/min on day 1 after ascompared with the controll, and these changes were closely related to the final temperattrre. At 10℃/min cooling rate the severity of injury of -40℃ sample was simitar to that of -20℃ with a rate of temperature drop of 1℃/min. After 3 days there was recovery of all the parameters. The results showed that VEC were injured even at 0℃ in the absence of crvoprotector, and the slower the cooling rate and the Lower the temperature, the severer injuries were induced.
