摘要
使用DEAE-52离子交换层析和ATP琼脂糖亲和层析方法,从人体肿瘤细胞株、大鼠肝脏和心脏、家兔和小鼠肝脏组织中纯化了主要的热应激蛋白—HSP70,用氨基酸自动分析仪分析它的氨基酸组成。结果发现:在所测的5种来源的HSP70中,含量位于前3位的氨基酸,除大鼠心脏为甘氨酸、苯丙氨酸和谷氨酸外,其余的均为甘氨酸、谷氨酸和天冬氨酸,所有来源的HSP70均富含苯丙氨酸、赖氨酸、缬氨酸、亮氨酸和丙氨酸。这些结果为探讨HSP70在劳动卫生学中的应用提供了参考数据。
The major heat stress protein HSP7o was purified from human leudaemia cancer cell line
K562, rabbit liver,rat liver, ratheart and mouse liver with two-step procedures of DEAE-52 ion
exchange chromatography and the affinity chromatography onATP-agaroea and their amino
acid composition was detemined with Amino Acid Analyser. The result showed that the first
threericliest amino acids in HSP70 of all origins tested were Glu Glu and Asp.except those of
rat heart(Gly.Phe and Glu).Lys, Phe,Val,Leu and Ala were very ricb in HsP70 of all origins. The
result proviued the referenced data for exploring the application ofHSP70 in labour hygiene and
occupational diseases.
