摘要
目的建立反相高效液相色谱法测定人血清中维生素A的方法,以评价人体维生素A营养状况。方法移取血清,加入乙腈乙醇混合溶液及正己烷进行萃取,旋涡混合、离心后,移取正己烷层氮气吹干,残渣用甲醇溶解后进样。色谱条件:XTerraTM RP18色谱柱,5μm,3.9 nm×150 mn,柱温:30℃,流动相为甲醇: 水(92:8);流速:1 mL/min;检测波长325nm。结果维生素A在25~800ng/mL浓度范围呈线性,R2=0.9993, 平均回收率为95.2%,RSD<7%,全部操作可在30min内完成。结论改进的检测人血清中维生素A含量的方法, 快速、灵敏、准确,简便易行。
Objective To establish a HPLC assay for the determination of vitamin A in human serum and evaluate the nutritional value of the body's vitamin A. Methods The serum was extracted with n-hexane after the protein in the serum was deposited with acetonitrile /alcohol. The layer of n-hexane was dried with nitrogen, and the leavings dissolved with mobile phase and determined by HPLC. Vitamin A was separated on XTerra?RP18 5μm, 3.9 mm× 150mm column, and detected at 325 nm with UV detector by using methanol/water(92:8) as mobile phase when the column temperature at 30℃. Results The range of calibration curve is 25 -800 ng/mL (R2=0.9993). The average extracted recovery is 95.2%, RSD<7%. The whole operation can be finished in less than 30 minutes. Conclusion The method is sensitive, rapid, accurate and simple.
