摘要
目的:研究MMP7和FasL在大肠癌的表达及其意义。方法:应用免疫组化SP法检测MMP7和FasL蛋白在正常黏膜、非瘤性息肉、腺瘤和大肠癌等4组不同大肠病变组织中的表达情况,分析其间的相互作用关系。结果:①MMP7蛋白染色阳性率在4组分别为8.3%,10.0%,52.6%和70.4%,后2组显著高于前2组(均P<0.01),并且第4组显著高于第3组(P<0.05);FasL蛋白在4组阳性率分别为:0、10.0%、34.2%和63.6%。大肠癌组中FasL蛋白的表达阳性率显著高于前3组(均P<0.01)。②MMP7和FasL蛋白在大肠癌的表达与患者的性别、年龄、部位无明显相关性(P>0.05),随着分化程度的降低有增加趋势(P<0.05),在淋巴结有转移组、Duke’s分期C期及以上组分别高于淋巴结无转移组、Duke’s分期A、B期组(P<0.05)。③MMP7蛋白阳性表达与FasL蛋白阳性表达正相关(P<0.01)。结论:MMP7与FasL在腺瘤和大肠癌均有高表达,在大肠癌的发病过程中,它们的共同高表达具有协同效应,共同促进大肠癌的发生和发展。
Objective: To investigate the expression of MMP-7 and FasL in colorectal carcinoma. Methods: Tissue specimens of normal colorectal mucosae, non-tumorous polyps, adenomas and adenocarcinomas were examined for MMP-7 and FasL with streotavidion-peroxidase(S-P) immunohistochemistry technique, and analyzed their interacting relationship. Results: ①The positive rates of MMP-7 protein expression in these 4 groups are 8.3%,10.0%, 52.6% and 70.4% respectively. The latter two groups are significantly higher than the former two respectively (P<0.01), and the positive degree in adenocarcinomae is significantly higher than adenomas(P<0.05);The positive rates of FasL protein expression in these 4 groups are 0、10.0%、34.2% and 63.6% respectively, the positive rate is significantly higher in adenocarcinoma group than it in others(P<0.01).②There is a tendency that poorly differentiated colorectal carcinomas have more membranous expression of MMP-7 and FasL. The positive rate in low differentiated group is higher than it in moderate and high differentiated group(P<0.05).And it in the C stage of Duke's group with lymph metastasis group is higher than the A, B stage of Duke's group without lymph metastasis group respectively (P<0.05). ③The positive rate of MMP-7 protein expression is positively correlated with the positive rate of FasL protein expression. Conclusion: MMP-7 and FasL are all highly expressed in adenoma and adenocarcinoma, they cooperate with each other during the carcinogenesis of colorectal carcinoma.
