摘要
目的 :研究安托可金在犬体内的药代动力学。方法 :采用石墨炉原子吸收法测定血浆中安托可金的浓度 ,对犬静脉注射后的药动学进行了研究。该测定方法的最低检测浓度为 0 .1μg/ml,线性范围为 0 .1~5 .0 μg/ml,回收率大于 90 %。 结果 :犬静注安托可金后符合二房室模型处置特征 ,分布半衰期为 0 .5 6~ 0 .72h ,消除半衰期为 18.1~ 2 2 .8h ,中央室分布容积为 0 .12 3~ 0 .15 4L/kg。在 5 ,10 ,2 0mg/kg的剂量下AUC分别为 4 0 .8,74 .1和 177.5 μg·h/ml。 结论 :安托可金给药剂量和AUC基本呈线性关系 ,提示安托可金在犬体内处置属于线性动力学 ,其药动学参数呈剂量非依赖性。
OBJECTIVE:To study the pharmacokinetics of 0325k_ 1-1 in dogs. METHOD:The concentration of 0325k_ 1-1 in the plasma of dogs was determined by flameless atomic absorption spectrophotometry. The minimal detectable concentration was 0.1 μg/ml, the linearity was between 0.1~5.0 μg/ml and the recovery was higher than 90%.RESULT:The plasma concentration-time profiles fitted to two compartments after iv administration, the distribution t _ 1/2α was 0.56 h to 0.72 h,the elimination t _ 1/2β was 18.1 h to 22.8 h,the distribution volume of central compartment V_ c was 0.123 L/kg to 0.154 L/kg. The AUC was linear with the dose that was injected. CONCLUSION:The Pharmacokinetics property of 0325k_ 1-1 in dogs is linear.
