卡铂的药物毒性及疗效与用药时间关系的实验研究 被引量:1

摘要 本研究观察光-暗周期下荷瘤小鼠应用卡铂后的生存时间、体重、白细胞计数、酸性α-萘酸脂酶染色阳性淋巴细胞计数及病理学改变,以证明卡铂毒性及疗效的时间节律性。结果表明,卡铂的最小毒性时间为动物睡眠早期,此时药物对骨髓的毒性较小,疗效较好。提示正确选择用药时间,可以降低卡铂的毒、副作用,更好地发挥其疗效。 he platinum-containing compounds has become amajor chemical agent in the treatment of cancer. A cir-cadian rhythm in the susceptibility of rodents and hu-man being to cisplatin has been demonstrated, the maxi-mal tolerance being found in the animal's active phase.Carboplatin is a second generation analog. Two studieswere performed on mice with carboplatin under 12: 12light dark cycle to study its chronotoxity and chronoef-fectiveness. In study I, single intraperitoneal injectionof 192mg/kg (LD_(50) carboplatin was given to fourgroups of mice at four different circadian stage. It wasfound that at 50% the overall mortality of mice, therewas a mortality difference of 28% for mice receiving thedrug at 9 a. m. to 71% for mice receiving drug at9 p.m. It demonstrated that carboplatin was bettertolerated in the animal's eraly sleep phase. In study II,S_(180) tumor-bearing mice were treated with 50mg/kg ofcarboplatin. The longest mean survival time and thelowest marrow toxity occured in the group which re-ceived the drug at the beginning of the sleep phase. Itshowed that the susceptibility of mice to carboplatin iscircadian stage dependent. These data clearly demon-strate that, by timing the administration of drugs ac-cording to body rhythms, such as the host susceptibili-ty-resistance rhythm to a drug, one can gain a therapeu-tic advantage over an approach which ignores suchrhythms.
