摘要
目的观察褪黑素对体外培养的人视网膜色素上皮细胞生长、增殖的影响,寻找药物以防治增殖性玻璃体视网膜病变。方法分别将不同浓度的褪黑素(0,125ng·L-1,250ng·L-1,500ng·L-1)加入体外培养的视网膜色素上皮细胞培养液,72h后用四甲基唑盐(MTT)比色法在自动酶标测定仪测540nm处的吸光值A,并计算出不同浓度条件下的细胞增殖抑制率。结果褪黑素在不同浓度下抑制视网膜色素上皮细胞增殖,并存在剂量-效应关系,实验组125ng·L-1和500ng·L-1组与对照组比较有显著性差异(P<0.001),但实验组250ng·L-1组与对照组之间的差异无显著性。结论褪黑素能抑制体外培养的人视网膜色素上皮细胞增殖,可作为防治增生性玻璃体视网膜疾病的侯选药物作进一步研究。
Objective To investigate the effect of melatonin on proliferation of cultured human retinal pigment epithelial cells (hRPECs), search for effective drug for the treatment of PVR. Methods Different concentration of melatonin (0,125 ng·L-1, 250 ng·L-1, 500 ng· L-1) were added to hRPECs respectively , after 72 hours using MTT method at automatic enzyme labeling machine value of A were calculated at 540 nm and inhibition rate of retinal pigment epithelial(RPE) cells were measured by MTT. Results Melatonin at different concentration could inhibit RPE cells in dose dependent manner, in comparision of different experimental group 125 ng·L-1 and 500 ng·L-1 with control group difference was highly significant(P< 0.001) ,but comparision of experimental group 250 ng· L-1 with control difference was not significant. Conclusion Melatonin can effectively inhibits hRPECs growth in vitro, and it might be a new candidiate drug of preventing for patients with proliferative vitreoretinopathy.
