Inactivation of SARS coronavirus in human plasma by methylene blue/light method

摘要 目的:探讨亚甲蓝/光化学法灭活人血浆中SARS冠状病毒(SARS CoV)的效果。方法:从SARS患者血清中分离出冠状病毒,经VeroE6细胞增殖后将病毒加入含有不同浓度亚甲蓝的人血浆中,再用亚甲蓝/光化学法处理不同时间,取样接种细胞,观察细胞病变作用。结果:在激发光源照度为40000lx条件下,经1μmol/L的亚甲蓝作用30min可灭活血浆中6.5lgTCID50/ml的SARS CoV,经5μmol/L的亚甲蓝作用3min可灭活上述滴度病毒。结论:亚甲蓝/光化学法能够灭活血浆中的SARS CoV。 Objective: To investigate the inactivation effect of methylene blue / light method on SARS-coronavirus (SARS-CoV) in human plasma. Methods: SARS-CoV was isolated from the serum of SARS patient and reproduced by VeroE6 cell line. After added to the human plasma which contained different concentrations of methylene blue, SARS-CoV was treated with methylene blue / light for different periods of time and then the sample of plasma was taken to inoculate the cell line for assay of the cytopathic effect. Results: About 30 minutes were needed to inactive SARS-CoV (6.5 lgTCID 50/ ml ) in the plasma which was exposed to visible light (40 000 lx) in the presence of 1 μmol / L of methylene blue and only 3 minutes were needed to inactive above titer of viruses if the concentration of methylene blue increased to 5 μmol / L. Conclusion: SARS-CoV in human plasma can be inactivated by methylene blue / light method.
