摘要
[目的]观察不同浓度的苦参碱对正常人肝细胞的影响。[方法]浓度分别为250mg/L、500mg/L和1000mg/L的苦参碱体外培养肝细胞6d,观察对照组和苦参碱组的细胞形态学改变,通过MTT法和细胞蛋白量检测(BCA)观察细胞存活情况,检测乳酸脱氢酶(LDH)观察细胞膜的完整性。[结果]500mg/L和1000mg/L组细胞在第6d出现明显形态学改变和大量细胞死亡;MTT检测值在500mg/L组第6d,1000mg/L组第4d和第6d与对照组比较有显著性差异(P<0.05),细胞蛋白量检测值在1000mg/L组第4d和第6d与对照组比较有显著性差异(P<0.05);在500mg/L组第2d和第6d及1000mg/L组第2、4、6d,LDH值比对照组明显升高,具有统计学意义(P<0.05)。[结论]苦参碱浓度在500mg/L^1000mg/L时,对人肝细胞有较明显的损害作用。
To investigate the effect of different concentrations of matrine on the human hepatocytes in vitro. The human hepatocytes were cultured respectively at 250mg/L, 500mg/L and 1000mg/L matrine for 6 days, the morphological changes of cells were observed. The alive cell number was detected by MTT assay and BCA assay. Content of LDH (lactate dehydrogenate) was measured to find out the integrity of the cell membrane. The cells cultured with 500mg/L and 1000mg/L matrine appeared distinct morphological changes and many dead hepatocytes were found in the sixth day. The values of MTT assay decreased in the fourth day of 1000mg/L group and in the sixth day of 500mg/L and 1000mg/L group, was statictically significant compared to that in the control group (P<0.05). The protein quantities of cells in the fourth and sixth days of 1000mg/L group, were significantly different compared to the control group (P<0.05). The value of LDH increased in the second and sixth day of 500mg/L group, and in the second, the fourth and the sixth days of 1000mg/L group, which was statistically significant compared to the control group(P<0.05). [Conclusion] When the concentration of matrine was 500mg/L~1000mg/L, it could damage human hepatocytes in vitro obviously.
