电针对更年期大鼠血中性激素水平和下丘脑P物质mRNA表达的影响 被引量:8

Effect of Electro-acupuncturing on Gonadal Hormone Level of Plasma and Substance P mRNA Expression of Hypothalamus in Climacteric Rats

摘要 目的:探讨更年期大鼠血中性激素的变化与下丘脑神经肽P物质(SP)mRNA表达的关系及电针足三里穴对其的影响。方法:将实验动物分为三组, 分别为正常对照组(INT)、更年期组(CLI)、更年期针刺组(CLI+EA)。用放射免疫分析方法(RIA)测定大鼠血中雌二醇(E2)、卵泡刺激素(FSH)的含量;HE染色观察大鼠卵巢形态。提取下丘脑RNA,用RT-PCR方法,获得下丘脑SPmRNA表达产物,琼脂糖凝胶电泳检测,计算机图像分析系统处理数据。结果:与对照组相比,更年期大鼠血中(E2)含量明显降低,血中FSH含量明显升高,下丘脑SPmRNA表达产物较正常对照组明显增强,电针“足三里”穴的更年期组大鼠血中E2含量较未电针组明显增加,血中FSH明显降低, 下丘脑SPmRNA RT-PCR产物光密度值较未电针组减少, 有显著性差异。结论:电针“足三里”穴改变更年期大鼠血中性激素水平可能与电针影响下丘脑SPmRNA的表达及生成有关。 Objective: To investigate the mechanism of the regulation for electro-acupuncturing on gonadalhormone level in plasma of the climacteric rats. Methods: Experimental rats were divided into three groups: intactgroup (INT), climacteric group (CLI) and climacteric and electro-acupuncture group (CLI+EA). Radioimmunoassay(RIA ) was used to measure the level of the estrogen (E2) and follicle stimulating hormone ( FSH) in the plasma, thechange of the morphology of the rat ovaries was observed by HE staining.The mRNA expression of SP (substanceP) and glyceraldehyde phosphate dehydrogenase(GAPDH) of rats hypothalamus was measured by the RT-PCRtechnique and tested by the method of agarose gel electrophoresis and results were processed with a computerizedimage analysis system. Results: Compared with rats of INT group, the plasma E2 level of rats of CLI groupsignificantly lowered while plasma FSH level significantly increased, the number of developing follicles in ovariesreduced and the expression level of SP mRNA of hypothalamus was higher. However, the plasma estrogen level inrats of CLI+EA group was higher and FSH level was lower than these of the CLI group; the SP mRNA expressionlevel of hypothalamus in rats of CLI+EA group was lower than that of the CLI group. Conclusion: The level of E2and FSH in plasma of climacteric rats could be regulated by electro-acupuncture of Zusanli acupoint and may affectthe expresssion and synthesis of SP mRNA in the rat hypothalamus.
