Fertilization by intractoplasmic sperm injection in goat oocytes

摘要  探讨了显微操作条件的改善对山羊卵母细胞胞质内显微受精(ICSI)的影响。结果表明,将精子操作液中聚乙烯吡咯烷酮(PVP)浓度从10%(10g/L)降低到8%和5%后,ICSI胚的卵裂率差异不显著,但正常卵裂率、桑葚胚率和囊胚率随PVP浓度的下降而上升;PVP浓度为5%和10%时,正常卵裂率分别为69.6%和58.5%(P<0.05),桑葚胚率分别为45.6%和35.2%(P<0.05),囊胚率分别为26.9%和16.9%(P<0.05),表明含5%PVP的精子操作液更有利于ICSI胚的发育。显微注射时将卵母细胞第一极体置于相当于时钟6点的位置与12点位置相比,ICSI胚的分裂率没有差异,而正常卵裂率分别为69.9%和65.4%(P>0.05),桑椹胚率分别为45.5%和42.6%(P>0.05),囊胚率分别为26.7%和20.4%(P>0.05),表明注射时极体置于6点的位置有利于ICSI胚的体外发育。 <Abstrcat>This study was conducted to investigate factors influencing ICSI in Goat.In experiment 1,three concentrations (10%,8% and 5%) of polyvinylpyrrolidone (PVP) in the sperm injection vehicle solution were compared.There was no effect of the three PVP concentrations on cleavage rate after sperm injection,but regular cleavage rate,morulae rate and blastocyst rate were improved with decreasing concentration of PVP,cleavage rate (69.6% and 58.5%),morulae rate (45.6% and 35.2%) and blastocyst rate (26.9% and 16.9%) were significantly higher with 5% than 10% PVP (P<0.05).5% PVP in the sperm injection vehicle solution could improve the development of oocytes after ICSI.In experiment 2,two positions (6 and 12 o'clock) of the first polar body (PB) located in oocyte at ISCI were examined.There were no significant differences in the cleavage rate,regular cleavage rate (69.9% and 65.4%,P>0.05),morulae rate (45.5% and 42.6%,P>0.05),blastocyst rate (26.7% and 20.4%,P>0.05) between the two positions of the PB,but they were improved at 6 o'clock of the polar body.Oocytes should be injected with the polar body at 6 o'clock.
