摘要
目的 研究酒精对大鼠胚胎脑星型胶质细胞热休克蛋白HSP70表达和细胞超微结构的影响。方法 对体外分离培养的大鼠胚胎脑星型胶质细胞分别施以不同剂量酒精 (1、5、2 5、5 0mmol L) ,以免疫细胞化学技术观察酒精对其HSP70表达的影响 ,并结合电镜技术观察其超微结构的变化。结果 随酒精剂量增加 ,胎鼠脑星型胶质细胞HSP70阳性表达与剂量成正相关 ;2 5mmol /L和 5 0mmol /L剂量组存活细胞个数减少 ,细胞膜和线粒体发生损伤 ,部分细胞出现凋亡和坏死。结论 提示酒精能通过增强星型胶质细胞HSP表达抑制维持正常功能蛋白质的表达 ,并可通过损伤星型胶质细胞的细胞膜和线粒体导致凋亡和坏死。
Objective\ To explore effects of alcohol on the expression of heat shock protein 70(HSP70) and su per microstructure of astrocytes. Methods Different dosage of a lcohol(1,5,25,50mm ol/L) was administered to astrocytes of rat embyos. Effects of alcohol on expre ssion of HSP70 were studied by immunocytochemistry, and changes of super microst ructure of astrocytes were observed by electron microscope.Results The results showed that with increase of alcohol dosage, the expression of HSP70 increased, hepatin granule and survival astrocytes in 25mmol/L and 50mm ol/L dosage group decreased, cell membrane and mitochondrion were damaged, and e ven apoptosis and necrosis occurred.Conclusion It suggested th at alcohol could increase expression of HSP of astrocytes, which could depress the expression of normal function protein, and mainly impaired cell membrane and mitochondrion, wh ich could lead to apoptosis and necrosis.
