表皮生长因子促进大鼠脑梗死后的早期神经功能康复 被引量:5

Exogenous epidermal growth factor promotes early neural function recover after experimental cerebral infarction in rats.

摘要 目的 观察脑室内注入表皮生长因子(EGF)对大鼠脑皮层梗死后神经康复的作用。方法 采用肾血管性高血压大鼠复制成一侧大脑中动脉皮层支闭塞(MCAO)模型。MCAO术后2 4~4 8h ,32只实验组大鼠侧脑室注入1 0 μLEGF( 0 1 μg/ μL) ,连续2天,共2 μg/ μL ,其余6 4只MCAO对照组大鼠只注入不含EGF的等量溶液或不给予任何处理。MCAO术后1、2、3和4周,行神经功能评估和脑切片,HE染色计算脑梗死灶大小,并与8只假手术对照组大鼠比较。结果 EGF组大鼠神经功能在MCAO术后1周和2周均较同期对照组大鼠改善更明显(P<0 .0 5) ,在MCAO术后3周和4周则无差异(P >0 . 0 5)。EGF组大鼠在MCAO术后各时期的脑梗死灶体积均较同期对照组大鼠为小(P <0. 0 5)。结论 EGF促进大鼠脑皮层梗死后早期神经功能康复,但对远期神经功能康复无明显影响(P >0 .0 5)。 Objective To observe neural function recover in adult rats with neocortical infarction by intraventricular administration of exogenous epidermal growth factor(EGF). Methods Twenty-four to forty-eight hours after the right middle cerebral artery occlusion(MCAO) in renovascular hypertensive rats, ten μL EGF(0.1 μg/μL) was infused consecutively into lateral ventricle for 2 days in 32 experimental rats(EGF group) and the same amount of vehicle or none in 64 MCAO rats as controls. At the end of 1,2,3, and 4 weeks after MCAO, the brains of eight rats from each group respectively were sectioned and stained with HE for infarct size determination after neurological function evaluation. Sham controls were matched at the baseline. Results Much better neurological function in EGF group than in vehicle group or in stroke control group was observed at the end of 1,2 weeks, but not different at the end of 3,4 weeks after MCAO. Infarct size was smaller in EGF group than in vehicle group or in stroke control group at the end of 1,2,3,4 weeks after MCAO. Conclusions EGF promotes early neural function recover after experimental cerebral cortex infarction in hypertensive rats.
