离心液相色谱法快速分离纯化神经节苷脂 被引量:2

摘要 神经节苷脂在脊椎动物的神经组织细胞膜上含量丰富,除有受体的功能外,还有许多其它功能。采用离心液相色谱法,以国产硅胶G-60为填料,分离纯化神经节苷脂,取得了满意的效果。经分离后产物的脂结合唾液酸(LBSA)含量,由14.3%提高到24.1%。以蛋白质含量为代表的含氮化合物由22.0%降至9.6%。回收率为90.25%。特点是操作简便、周期短、成本低、效果好。优于latrobeads柱层析法。 A method for isolation and purification of Gls by CLC was recommended. Gangliosides are present in neuronal cell membrane of vertebrate in high concentration.They appear to be receptors and with other function.We had isolated and purified the Gls by CLCmethod in which we utilized silica gel G-60 as a filter. We found that the content of LBSA inthe crude Gls was improved from 14. 3% (w/w) to 24. 1% after CLC and the nitrogen compounds (calculated as protein) were decreased from 22. 0% to 9. 6% (w/w). The rate of recovery was up to 90. 25%. So, this method is better than the latrobeads column chromatography. It is less in demand of solvent than the latrobeads method.
