急性实验性脑外伤后脂质过氧化反应及其超微结构变化的动态观察

摘要 目的通过对脑损伤灶周边部位脑组织超微结构的动态观察,进一步阐明脂质过氧化反应与继发性脑损害的关系。方法在建立大鼠脑外伤模型的基础上,测定脑组织及血液中MDA和SOD含量,动态观察脑组织在光镜和电镜下的病理学改变。结果①脑组织与血液中的有关指标具有一定规律性和相关性;②损伤灶周边部位的脑组织呈明显的继发性病理改变,并具可逆性变化;③伤后6h脑组织中MDA含量的增加和SOD性的下降最明显,此时脑组织病理损害程度也最重;④伤后脑组织MDA含量无明显变化时损伤灶周边部位的脑组织却已发生了明显的病理损害。结论脂质过氧化反应只是参与和加重了脑组织的继发性病理损害,但在脑组织最早期的继发性损害中,可能有其它更重要的因素在自由基之前起作用,且细胞毒性水肿出现得更早。 Purpose The brain ultramicro structure of the damaged periphery was dynamically observed in order to further clarify the relations between the reaction of lipoperoxidize and the secondary damage in rats brain. Methods Experimental acute brain injury were made in rats. The contents of lipoperoxide(LPO,MDA)and superoxid dismutase(SOD) in brain tissue and in blood were examined after the brain damage, and the examidnation of the pathophysiological changes were done dynamically under light and electric microscope. Results ①There were certain regularity and correlation in relational index between brain and blood.②The secondary pathophysiological changes were showed in the edge of the damaged reglon. ③the contents of MDA did significantly increase in brain after damage 6hrs,but SOD droped. In that moment the pathophysiological damage in brain were the gravest. ④The pathophysiological damages were significantly showed in the edge of injuried brain, . but the contents of MDA did not change obviously at the same time. Conclusions The lipoperoxidized reaction only joined and exacerbated the secondary damage of brain tissue after injury. There were perhaps any more important factors roling in this pathophysiological damage before the oxygen radicals did,and the cell tokicity dropsy was regarded appearing more early.
