摘要
目的 研究黄芪丹参复方成分提高胎盘血供的分子机制,为临床有效防治胎盘血供不足所致妊娠并发症提供思路。方法 提取分离黄芪、丹参有效复方成分,运用一氧化氮合酶(NOS)阻滞剂L-精氨酸甲酯(L-NAME)制作一氧化氮合成阻滞大鼠模型,黄芪丹参复方成分妊娠同步干预,核酸原位杂交技术检测妊娠18 d胎盘滋养细胞内皮型一氧化氮合酶(eNOS)、血管内皮生长因子(VEGF)mRNA表达变化。结果 中药治疗组、对照组、空白组eNOS mRNA表达明显高于模型组(P<0.01)。一氧化氮合成阻滞模型VEGF mRNA表达与空白组、中药治疗组、硝酸甘油治疗对照组相比均明显降低(P<0.01)。结论 黄芪丹参复方成分可能提高胎盘滋养细胞NO合成与释放入循环,促进胎盘血管网络形成,从而维持妊娠期胎盘循环的高流速、低阻力状态,达到有效防治妊娠高血压综合征及胎儿宫内生长迟缓等胎盘血供不足类疾病的目的。
Objective To supply clinical thoughts on effectively preventing and treating pregnant complications caused by insufficient placental blood supply, by exploring the molecular mechanism of the components of Mongolian milkvetch root-Danshen root compound elevating placental blood supply. Methods Separate and abstract the effective components of Mongolian milkvetch root-Danshen root, apply L-Arginine methyl ester (L-NAME) to pregnant rats to make NOS blocking model with the interference of the components of Mongolian milkvetch root-Danshen root compound in pregnancy and then inspect the change of the expression of placental nourish cells eNOS mRNA and VEGF mRNA on the day 18 of pregnancy by the technology of in situ hybridization of nucleic acid. Result The expression of eNOS mRNA of treat group by Chinese medicine, contrast group and blank group were obviously higher than that of model group (P<0.01). The expression of VEGF mRNA of model group were obviously lower than that of blank group, treat group by Chinese medicine and contrast treat group by nitroglycerin (P<0.01). Conclusion The components of Mongolian milkvetch root-Danshen root compound can improve the speed of the composition of NO of placental nourish cells and the speed of releasing NO into circulation, accelarate the form of placental vas and keep placental circulation in the state of high speed and low resistence. In the way of above, it can do well in preventing and treating such diseases caused by insufficient placental blood supply as PIH and intrauterine growth retardation (IUGR).
