摘要
利用田间自然诱发方法对100余份甘薯品种连续进行3年茎线虫病的抗性鉴定,选取其中21份抗、感性稳定的品种作为供试材料,含高抗品种10个、抗病品种2个、感病品种2个、高感品种7个.以徐781和栗子香的总DNA为模板,从70个随机引物中选出16个引物对其进行随机扩增,均得到了稳定的RAPD图谱.扩增出的片段分子量为0.3~2.0 kb,条带数在1至9之间,共扩增出119个条带,其中,84.03%的谱带为多态性带.利用NTSYS-PC软件中的Simqual程序估算DNA的相似系数,UPGMA程序构建遗传相关树状图.遗传多态性的RAPD分析可为甘薯抗茎线虫病品种选育的亲本组配提供依据.
More than 100 sweetpotato varieties inoculated with stem-nematodes in the field were evaluated subsequently for resistance to stem-nematode under field conditions from 2001 to 2003. Of the 21 chosen varieties, according to the identification, 10 were highly resistant, 2 resistant, 2 susceptible and 7 highly susceptible. Sixteen primers, screened from 70 total DNA of Xu781 and Lizixiang, were used to amplified the total DNA of these genotypes. The result showed that each primer provided a clear and stable RAPD map; the length of the products ranged 0.3—(2.0 kb); and the numbers of amplified bands was from 1 to 9. The polymorphic bands accounted for 84.03% of the total 119 amplified bands. Similarity coefficient was calculated by Simqual of NTSYS-PC. Dendrogram was built by UPGMA. RAPD analysis of genetic diversity would be useful in cross combination setting of breeding for high resistance to stem-nematode in sweetpotato.
