摘要 目的 :以 2 ,4 二硝基氯苯 (DNCB)建立小鼠溃疡性结肠炎 (ulcerativecolitis,UC)模型并探讨溃疡性结肠炎与一氧化氮 (NO)、肿瘤坏死因子 a (TNFα)的关系。方法 :将小鼠随机分为实验组和对照组 ,用 3 3%DNCB涂腹致敏 ,再以 0 1%DNCB、 0 2 %DNCB、 0 4 %DNCB (对照组以 6 0 %乙醇 )灌肠 ,1次 /d ,连续 4d。观察疾病活动指数、结肠大体和镜下改变。采用硝酸还原酶法和生物素链亲和素ELISA法测定血清NO和TNFα。结果 :0 2 %DNCB、 0 4 %DNCB组与对照组相比 ,疾病活动指数、组织病理学评分以及NO、TNFα 活性均有显著升高 (P <0 0 5或P <0 0 1)。结论 :DNCB能够诱导小鼠溃疡性结肠炎 ,NO、TNFα 参与溃疡性结肠炎的病理过程。 Objective: To estabilish mouse ulcerative colitis model of ulcerative colitis (UC) with an allergen 2,4-dinitrochlorobenzene (DNCB),and observe the relationship between NO、TNF α and ulcerative colitis.Methods: BALB/c mice were randomly divided into control (60%ethanol) and DNCB groups. After they were sensitized by smearing 3.3%DNCB on the abdominal skin,they received intracolonicly DNCB of 0.1%,0.2% and 0.4% DNCB respectively once a day. The weight,stool viscosity and hematochezia was observed and accumulated as disease active index (DAI) score. The pathological changes in colon tissue were judged macropathologically and by means of microscope.NO and TNF α activity was determined. Results: There are significant rise in DAI,pathologic score and NO and TNF α activity in 0.2%DNCB、0.4%DNCB groups as compared with control group. Conclusion: Mouse colitis could be induced by DNCB,and NO 、TNF α involved in the pathological process of the disease .
