摘要
目的 研究Survivin在白血病中表达的临床意义及其在白血病细胞凋亡中的作用。方法 应用蛋白印迹法检测1 8例白血病患者和 1 0例正常对照组患儿外周血中单个核细胞Survivin的表达 ;采用免疫组化法测定Fas和Caspase表达 ;运用四格表的确切概率法进行Survivin表达与白血病患者临床特点和Fas、Caspase表达的进行相关性分析。结果 1 .白血病患者1 8例中 1 3例Survivin表达阳性 (72 .2 % ) ,正常对照组 1 0例Survivin检测均为阴性。Survivin表达在不同白血病类型、WBC组间有显著差异 (P <0 .0 5)。 2 .白血病患者 1 8例中 ,9例Fas表达阳性。经统计学分析 ,Survivin表达与Fas表达无相关性(P =0 .1 1 6)。 3 .白血病患者 1 8例中 5例Caspase 3表达阳性 ,两者比较 (P =0 .0 0 2 ) ,有显著性差异。结论 Survivin基因可选择性的在白血病患者外周血单个核细胞中表达 ,而在对照组中无Survivin表达 ,表明Survivin表达与白血病的发生有一定关系。Survivin表达情况可能成为白血病临床诊断、预后判断的有效指标。Survivin的作用位点可能不在Fas水平 ,而主要通过抑制Caspase 3表达 ,在凋亡途径的下游发挥抗凋亡作用。
Objective To explore the relation ship between expression of survivin gene in leukemia cells and its clinical effects, and to study the mechanism of survivin resist-apoptosis function in leukemia.Methods Survivin expression was detected by Western blots analysis and expressions of Fas and Caspase were examined by immunohistochemistry in 18 leukemia patients.Results Thirteen cases in peripheral blood mononuclear cell survivin positive expression was detected in 18 leukemia patients(72.2%), but no survivin expression in 10 normal persons. There were significant difference of survivin expression in ALL/ANLL patients groups and different WBC groups(P<0.05)nine cases positive expression of Fas was examined in 18 cases leukemia. There was no correlation between survivin and Fas expression.Five cases positive expression of Caspase was examined in leukemia cases.There was significant difference between Survivin and Caspase expression.Conclusions Survivin gene express selectively in leukemia cell,but not in normal peripheral blood mononuclear cell, it hint that there is relationship between Survivin expression and leukemia happening; Survivin expression would be a effective monitoring index in diagnosing and prognosis criterion of leukemia. Survivin gene doesn't incude Fas expression, one path that Survivin gene which resist cell apoptosis is restraint Caspase expression.
