摘要
目的研究地塞米松和甲基强的松龙玻璃体内大剂量注射对视网膜的毒性。方法成年新西兰大白兔8只分为2组,第一组玻璃体腔内分别注射地塞米松磷酸钠注射液0.3 ml(5 mg/ml)和生理盐水0.3 ml作为对照。第2组注射用甲泼尼龙琥珀酸钠0.3 ml(40 mg/ml)(甲基强的松龙),另一眼注入甲基强的松龙溶剂0.3 ml(苯甲醇9 mg+注射用水1 ml)作为对照。注射前后分别行视网膜电流图检查,注药后1 d 和7 d 光镜观察视网膜切片。结果地塞米松造成视网膜 Müller 细胞肿胀和 Müller 纤维断裂.ERG b 波下降。甲基强的松龙造成视网膜全层结构破坏,坏死,ERG 波形消失。结论地塞米松和甲基强的松龙大剂量误注入眼内对视网膜有损伤,甲基强的松龙对视网膜的毒性更强,其溶剂对视网膜无明显毒性。
Purpose To evaluate the retinal toxicity after high dose of dexamethasone sodium phosphate injection and methylprednisolone sodium succinate sterile power intravitreal injection.Methods 8 New Zealand rabbits were devided into two groups:In one group dexamethasone sodium phosphate injection 0.3 ml (5 mg/ml) was injected intravitreally into eye and saline 0.3ml into the other eye as control.In the other group methylprednisolone sodium succinate sterile power 0.3 ml (40 mg/ml) was injected into one eye and methylprednisolone sodium succinate sterile power dissolvent 0.3ml (Benzyl alcohol 9 mg plus distilled water 1 ml) into the other eye as control.They were tested by ERG before and after injection.Retinas were observed by light microscope after 1 day and 7 days of injection.Results Müller cells edema and Müller fibers breaks were found after dexamethasone injection.B wave of ERG declined.Whole layer of relina broke deslroyed by neorosis after methylprednisolone sodium succinate sterile power injection.Ware of ERG disappeolred.Conclusion Retinal toxicity was found after high dose of dexamethasone and methylprednisolone sodium succinate sterile power intravitreal injection. Methylprednisolone sodium succinate sterile power was more toxic than dexamethasone.
