摘要
目的 :研究抗孕激素米非司酮 (mifepristone ,MIF)对人卵巢癌 3AO细胞株裸鼠移植瘤生长的抑制作用并探讨其作用机制。方法 :随机将 14只人卵巢癌细胞 3AO移植瘤荷瘤裸鼠分为两组 ,MIF组皮下注射MIF 0 .2mg/d ,对照组注射等量溶剂。观察治疗前后移植瘤的体积和重量。应用末端转移酶介导的生物素标记脱氧尿苷三磷酸缺口标记法 (TUNEL)和流式细胞术 (FCM)检测细胞凋亡率和肿瘤细胞周期时相的变化 ;免疫组织化学染色法检测移植瘤组织增殖细胞核抗原 (PCNA)和孕激素受体 (PR)的表达。光镜和电镜观察移植瘤细胞凋亡的超微结构变化。结果 :治疗 4周后 ,MIF组和对照组裸鼠移植瘤体积和重量分别为 1186 .14± 4 96 .14mm3和 1.30± 0 .4 3g ,2 199.85± 5 41.4 9mm3和 2 .30± 0 .31g(P <0 .0 1)。TUNEL测定显示 ,MIF组细胞凋亡率为 (11.33± 1.5 3) % ,对照组为(2 .6 7± 0 .5 8) % ,差异有显著性 (P <0 .0 5 )。FCM测定 ,MIF组凋亡率为 (15 .6 2±3.87) % ,也明显高于对照组的 (7.5 9± 1.4 7) % (P <0 .0 5 ) ,肿瘤细胞出现典型的细胞凋亡形态变化 ,并阻滞于G0 ~G1期 ,S期细胞显著减少 (P <0 .0 5 )。MIF组肿瘤细胞PCNA及PR表达率分别为 2 0 .5 %和 8.0 % ,明显低于对照组的 4 8.0 %和 2 2 .0 % (P <0 .0 5 )
Objective:To investigate the inhibitory effect and mech anism of ant iprogestins mifepristone on the xenograft of nude mice of human ovarian carcino ma 3AO cell line.Methods:14 nude mice with the xenograft of hu man ovarian carcinoma 3AO cell line were randomly divided into 2 subgroups,mifepristone group:0.2mg of mi fepristone was injected daily for 4 weeks,control group: vehicle oil was inject ed. The size and weight of xenograft were measured in the pre- and post-treatm en t. The apoptosis rate and cell cytokinetic phase of the xenograft implants were measured by TUNEL and flow cytometry(FCM).The changes of morphological and ultra structure were observed by electron microscopy.PCNA and progesterone receptor(PR ) expression were determined by immunohistochemistry staining method.R esults:After 4 weeks of treatment, the size and weight of the xenograft s of MIF group w ere 1186.14±496.14mm3 and 1.30±0.43g, which was lower than that of control g roup of 2199.85±541.49mm3 and 2.30±0.31g (P<0.01).TUNEL dem onstrated that the apoptosis rate of mifepristone group was(11.33±1.53)% whi ch decreased significantly than that of (2.67±0.58)% of control group( P<0.05).FCM also revealed t hat the apoptosis rate of mifepristone group was higher than that of co ntrol grou p,they were(15.62±3.87)% and (7.59±1.47)%,respectively(P<0.05). 3A O cell line of mifepristone group appeared a typical morphology changes and the form ation of apoptosis bodies, and was blocked at G 0-G 1 phase,and the radio of S pha se cells reduced significantly (P<0.05). The expression rates o f PCNA and PR of m ifepristone group were 20.5% and 8.0%, which were lower than that of 48.0% and 22 .0% of control group, respectively (P<0.05).Conclusion: Mifepristone significantly inhibites the proliferation activity of the xenografts of human ovarian cance r 3AO cell line in nude mice by inhibiting proliferation and inducing apoptosis of tumor cells.
