摘要
目的 :研究白细胞介素 - 2 (IL - 2 )对大鼠子宫平滑肌细胞增殖的影响 ,并探讨其可能的机制。方法 :以培养的大鼠子宫平滑肌细胞进行了以下实验 :以MTT比色法 ,测定IL - 2对该细胞增殖的剂量效应 ;用流式细胞技术 ,观察IL - 2对该细胞细胞周期各阶段DNA含量的影响 ;用激光共聚焦显微镜技术 ,检测IL - 2对该细胞内Ca2 + 水平的影响 ;用RT -PCR技术 ,检测IL- 2R在该细胞的表达。结果 :IL - 2 (10～ 10 0 0U/ml)可显著促进大鼠子宫平滑肌细胞的增殖。用流式细胞技术检测细胞中DNA的含量发现 ,IL - 2 (10 0U/ml)可提高该细胞由G1期进入S期的比例。激光共聚焦法检测到IL - 2 (10 0U/ml)可升高细胞内的Ca2 + 浓度。RT -PCR技术检测到细胞表达IL - 2RαmRNA、IL - 2RβmRNA和IL - 2RγmRNA三条链。 结果 :IL - 2参与了对大鼠子宫平滑肌细胞增殖的调节 ,其促进增殖作用可能与该细胞周期DNA含量的变化、Ca2 + 浓度的改变和IL - 2R的表达有关。
Objective:To investigate the effect o f IL-2 on the proliferation of rat uterin smooth muscle cells and explore its possible mechanism. Methods: Enha ncement effect of IL-2 on proliferation of cultured rat uterin smooth muscle cel ls, the effect of IL-2 on DNA content at various periods of cell cycle and on Ca 2+ concentration in the cells and the expression of interleukin-2 receptor s were detected or observed by MTT colorimetry, flow cytometry, laser scanning confoca l microscope and RT-PCR, respectively. Results:I L-2 of 10 to 1 000 U/ml signific antly stimulated the proliferation of cultured rat uterin smooth muscle cells. I L-2(100 U/ml) increased the ratio of rat uterin smooth muscle cells in G 1 and S phas es and increased the Ca 2+ release from rat uterin smooth muscle cells. Conclusion:IL-2 significantly enhanced the proliferation of rat uterin smooth muscle ce lls. The enhancement effect of IL-2 on rat uterin smooth muscle cell proliferati on is possibly related to the altering of DNA content, increased Ca 2+ conc entration and expression of interleukin-2 receptors in the cells.
