摘要 目的 :探讨洛伐他汀 (Lovastatin ,LOV)对HL - 6 0细胞凋亡的影响并观察细胞超微结构变化。方法 :以 4、8、16 μmol/LLOV处理HL - 6 0细胞 1~ 3d后 ,经DNA梯形带检测、DNA片段化百分率检测以及透射电镜等技术方法 ,观察细胞凋亡效应的发生。结果 :LOV处理HL - 6 0细胞后 ,有凋亡特征性的DNA梯形带出现 ;DNA片段化百分率增高 ;透射电镜下 ,HL - 6 0细胞呈现凋亡性变。结论 :LOV能诱导HL - 6 0细胞凋亡并使细胞超微结构发生凋亡性变。 Objective:To investigate the effects of cholesterol inhibitor Lovastatin(LOV) on cell apoptosis and to observe the changes of ultrastructure in HL-60 cells. Methods: After being treated with LOV at dosages of 4, 8, 16 μmol/L for 1 to 3 days,the cell apoptosis was measured with DNA ladder and DNA fragmentation rate assay,th e changes of ultrastructure in HL-60 cells were observed with the electron micro scope. Results:The DNA ladders were detectable and th e DNA fragmentation rate increased in LOV-treated cells;the morphological apoptosis c hanges were observed by transmission electron microscopy. Conclusion: Lovastatin is able to induce apoptosis and change the ultrastructure in HL-60 cells.
