摘要
目的 研究通脉降脂胶囊 (TMJZ)对四氯化碳所致小鼠急性肝损伤的保护作用及可能机制。方法 小鼠分别予TMJZ 1 50 ,75mg (kg·d)灌胃 (ig) ,连续 1周作预处理 ,然后用四氯化碳造成小鼠急性肝损伤 ,2 4h测定肝脏指数 (liverindex ,LI) ,血清谷丙转氨酶(serumglutamicpyruvictransaminase,SGPT)和肝组织谷丙转氨酶 (liverglutamicpyruvictransaminase ,LGPT)、甘油三酯 (triglyceride,TG)、总胆固醇 (totalcholesterol,TC) ,丙二醛 (malonaldehyde,MDA)、超氧化物歧化酶 (superoxidasedismutase,SOD)以及谷胱甘肽 (glu tathione,GSH)等指标 ,并观察肝脏组织学变化。结果 与四氯化碳模型组比较 ,经TMJZ预处理后SGPT、LGPT、TG、TC和MDA含量均明显降低 ,而GSH含量有所升高 ,同时SOD活力显著增强 ,并且TMJZ能明显减轻肝组织的病理变化。
Objective To study the hepatoprotective effect and possible mechanism of TongMaiJiangzhi Capsule(TMJZ) on experimental liver injury in mice. Methods In TMJZ groups, mice were orally treated with TMJZ at 150,75mg/(kg·d) respectively for 1 week. Then experimental acute hepatic impairment models were induced by carbon tetrachloride. After 12 hours, body weight and liver weight of mice were weighed. The level of GPT in serum and liver tissue and malonaldehyde(MDA), superoxidase dismutase(SOD), glutathione(GSH), triglyceride(TG), total cholesterol(TC) in liver were determined. Hepato-pathohistologic changes of mice were observed with light microscopy. Results TMJZ could inhibit the rising of serum glutamic pyruvic transaminase(SGPT), liver glutamic pyruvic transaminase(LGPT), liver MDA, TG, TC and liver index in CCl4 induced acute liver injury mice. They could also raise the content of liver SOD and GSH, and could reduce the liver lesion by CCl4. Conclusion It is revealed that TMJZ could prevent CCl4 from inducing acute liver injury.
